Human Sirtuin 4 ELISA Kit

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of human SIRT4 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. Standards and samples are captured by an antibody specific for SIRT4 pre-coated onto a 96-well microtitre plate, with detection via a biotinylated anti-SIRT4 antibody, an HRP-avidin conjugate and a TMB substrate. Absorbance is read at 450 nm on a colorimetric microplate reader.

The quantitative sandwich assay reports a standard curve range of 0.156–10 ng/ml and a sensitivity of 0.051 ng/ml, with a total assay time of 4 h 30 min on a pre-coated microplate configured as 12 × 8 well strips. Spiked recovery (n=5) averaged 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Linearity was assessed at sample dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma.

Supplied components include a pre-coated 96-well microplate, lyophilised standard (2 vials), detection solution A (120 µl), detection solution B (120 µl), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers (5 adhesive strips), with storage per component at +4°C or –20°C as instructed.
