The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and is read on a colorimetric microplate reader at 450 nm.

An antibody specific for RANTES is pre-coated onto the plate; standards and samples are added, washed, and then incubated with a Biotinylated Anti-RANTES Antibody followed by HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of stop solution, with the density of yellow proportional to the amount of RANTES captured in each well. Total assay time is 4h 30m. The stated product range is 0.156-10 ng/ml, with sensitivity of 0.073 ng/ml.

Reported recovery (n=5) was 80%-102% (average 91%) in serum, 81%-101% (average 91%) in EDTA plasma, and 80%-89% (average 84%) in Heparin plasma.
