Manufactured by antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human platelet derived growth factor subunit B (PDGF B) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, and is read colorimetrically at 450 nm in a microplate reader.

Quantitative measurement is achieved via a sandwich enzyme immunoassay in which standards and samples are captured by an immobilised PDGF B-specific antibody, detected with a biotinylated anti-PDGF B antibody, and visualised using an HRP-avidin conjugate and TMB substrate. The product range is 31.2–2000 pg/ml with a sensitivity of <11.3 pg/ml, and the total assay time is 4 h 30 min. Reported recoveries are 80–102% (average 91%) in serum (n=5), 81–100% (average 90%) in EDTA plasma (n=5), and 80–89% (average 84%) in heparin plasma (n=5). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is reported as 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (all n=5).

Kit components include the pre-coated 96-well microplate, two lyophilised standard vials, detection solution A (120 µl), detection solution B (120 µl), 30× wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and five adhesive plate sealers. Components are stored at +4 °C or −20 °C as indicated, and the kit should not be used past its expiration date.
