Human Paraoxonase 1 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of human Paraoxonase 1 (PON1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for PON1; standards and samples are added to the wells and bound PON1 is detected using a Biotinylated Anti-PON1 Antibody followed by HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue product that turns yellow on addition of stop solution, and absorbance is read at 450 nm on a colorimetric microplate reader.

The kit has a quantitative product range of 3.12–200 ng/ml and a sensitivity of <1.44 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery has been validated in serum (80–102%, average 91%; n=5), EDTA plasma (81–100%, average 90%; n=5) and heparin plasma (80–89%, average 84%; n=5), and dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across these sample types. Kit components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
