Human Mucin 5 Subtype B ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is designed for the in vitro quantitative determination of human MUC5B in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. An antibody specific for MUC5B is pre-coated onto a 96-well microtiter plate; standards and samples are added, followed by a biotinylated anti-MUC5B detection antibody and an HRP-avidin conjugate. After incubation and washing, TMB substrate produces a blue colour that turns yellow upon addition of stop solution, with absorbance read at 450 nm.

The assay has a working range of 0.625–40 ng/ml and a sensitivity of <0.237 ng/ml, with a total run time of 4 hours 30 minutes. Reported recoveries are: serum (n=5) range 83–100%, average 91%; EDTA plasma (n=5) range 90–99%, average 94%; heparin plasma (n=5) range 91–101%, average 96%. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 90–101%, 80–108%, 84–95% and 92–104% for serum; 82–105%, 89–91%, 87–109% and 74–104% for EDTA plasma; and 90–94%, 87–108%, 86–101% and 90–101% for heparin plasma.

Kit contents include a pre-coated microplate of 12 x 8 well strips, lyophilised standard (2 vials), detection solutions A and B (120 µl each), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers (5 adhesive strips). Components should be stored at +4°C or −20°C as instructed and the kit must not be used past its expiration date.
