Human Interleukin-11 ELISA Kit

The assay applies a quantitative sandwich immunoassay technique. The microtiter plate provided has been pre-coated with a monoclonal antibody specific to IL-11. Standards or samples are added to the appropriate microtiter plate wells and incubated. After washing to remove unbound IL-11 and other components, a biotin-conjugated polyclonal antibody specific to IL-11 is added; if present, IL-11 is captured by the pre-coated antibody and "sandwiched" by the biotin conjugate. Avidin conjugated to Horseradish Peroxidase (HRP) is then added to each well, followed by a TMB (3,3'5,5' tetramethyl-benzidine) substrate solution. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution, and the colour change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The sandwich ELISA can detect both natural and recombinant human IL-11 and has shown no cross-reactivity with various other cytokines super-family proteins.

Key specifications include a sensitivity of < 4 pg/ml and an assay range of 4–1600 pg/ml. Detection is colorimetric on a pre-coated microplate platform (12 x 8 well strips), with an assay time of 3h 30m. The kit is shipped and stored at 4°C, and includes two calibration diluents (Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant testing) for standard curve generation.
