Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Hemoglobin Subunit beta/ba1 in serum, plasma, or erythrocyte lysates. The assay employs an antibody specific for Hemoglobin Subunit beta/ba1 pre-coated onto a 96-well microtiter plate, and standards and test samples are added so that the target analyte present in each sample is bound by the immobilised capture antibody.

Following incubation and washing, a Biotinylated Anti-Hemoglobin Subunit beta/ba1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of target captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

Sensitivity is reported as less than 0.45 µg/ml, with an assay range of 1.56–100 µg/ml. The total assay time is approximately 4 hours 30 minutes. The platform is a pre-coated 96-well microplate arranged as 12 x 8 well strips, and kit components are stored at +4°C or −20°C as instructed. Recovery performance has been characterised, with average values of 91% in serum (range 80%–102%), 90% in EDTA plasma (range 81%–100%), and 84% in heparin plasma (range 80%–89%).
