Manufactured by Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of human Angiotensin I in serum, plasma or other biological fluids. The kit employs the competitive enzyme immunoassay technique, in which Angiotensin I present in samples or standards competes with a fixed amount of immobilized Angiotensin I on a pre-coated microplate for binding sites on a biotinylated anti-Angiotensin I antibody. Following incubation, unbound antibody is removed by washing and an HRP-Avidin conjugate is added. TMB substrate solution is then used to visualize the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The absorbance is read at 450 nm in a microplate reader, with colour intensity inversely proportional to the Angiotensin I concentration of the sample.

The assay covers a working range of 15.6–1000 pg/ml with a sensitivity of <4.31 pg/ml, and the total assay time is 2 hours 30 minutes. Recovery averages 92% in serum (range 85%–100%, n=5), 94% in EDTA plasma (range 85%–103%, n=5), and 93% in heparin plasma (range 87%–99%, n=5). Dilution linearity has been validated from 1:2 to 1:16 across serum, EDTA plasma and heparin plasma. Kit components should be stored at +4°C or –20°C as instructed, and the kit must not be used past its expiration date.
