Description

Optimized for real-time quantitative PCR, this 5x probe-based master mix from Solis BioDyne contains all components necessary to perform qPCR, with the exception of template, primers, and probe. The formulation includes HOT FIREPol® DNA Polymerase in a proprietary reaction buffer designed to enable detection of low copy number targets, and is specifically optimized for DNA hydrolysis probes based on 5' flap endonuclease activity. The polymerase is activated by a 12 min incubation step at 95°C, which prevents extension of non-specifically annealed primers and primer-dimers formed at low temperatures during reaction setup.

Mix composition includes HOT FIREPol® DNA Polymerase, 5x Probe qPCR buffer with 15 mM MgCl2 (yielding 3 mM MgCl2 in 1x solution), dNTPs, and BSA. Documented applications include detection and quantification of DNA and cDNA targets, gene expression profiling, microbial detection, and viral load determination. The recommended cycling protocol involves an initial activation at 95°C for 12 min, followed by 40 cycles of denaturation at 95°C for 15–20 s and annealing/extension at 60°C for 60 s. Reaction setup can be performed at room temperature, and it is recommended to work under laminar flow or in a PCR box to prevent contamination. Recommended template inputs are 0.1 pg/µl–10 ng/µl for cDNA and 10 pg/µl–4 ng/µl for gDNA.

Routine storage is at -18°C to -28°C, and shipping or temporary storage for up to 1 month at room temperature has no detrimental effects on product quality. The product is manufactured by Solis BioDyne in compliance with ISO 9001 and ISO 13485 certified Quality Management Systems, and is supplied for research use only. FIREPol® is an EU registered trademark of Solis BioDyne OÜ, headquartered in Tartu, Estonia.
