HOT FIREPol® Multiplex qPCR Mix

This 5× probe-based qPCR master mix from Solis BioDyne is optimised for highly sensitive and accurate multiplex quantification of up to four DNA or cDNA targets in a single real-time PCR reaction. It is designed for use with DNA hydrolysis probes, exploiting the 5′ flap endonuclease activity of the included HOT FIREPol® DNA Polymerase, which is activated by a 10 minute incubation at 95 °C to suppress non-specific amplification and primer–dimer formation.

The ready-to-use mix supplies HOT FIREPol® DNA Polymerase, a 5× Multiplex qPCR buffer, ultrapure dNTPs (including dUTP) and 15 mM MgCl₂ (3 mM in a 1× reaction). The presence of dUTP permits optional UNG treatment to prevent carryover contamination, although UNG must be purchased separately. Reaction set-up can be performed at room temperature, and the product offers up to 30 days of room-temperature stability, making it well suited to high-throughput workflows. It is robust with GC-rich templates and is intended for applications such as DNA and cDNA detection and quantification, gene expression profiling, microbial detection and viral load determination.

Recommended cycling starts with 95 °C for 10 minutes for polymerase activation, followed by 40 cycles of 95 °C denaturation (15–20 s) and 60 °C annealing/extension (60 s). Routine storage is at –18 °C to –28 °C; shipping and temporary storage for up to one month at room temperature does not affect product quality.
