Description

A serum-free differentiation medium from STEMCELL Technologies, designed to drive human liver organoids toward mature hepatic cell types. Used as part of the HepatiCult™ Organoid Kit (Human) workflow, it is applied after cultures have been expanded in HepatiCult™ Organoid Growth Medium (OGM), typically following 2–3 passages in OGM. Upon switching, organoids adopt a compact, dense morphology with thickened epithelia and show reduced expression of the stem cell marker LGR5 and the ductal markers KRT19, KRT7, and SOX9, alongside increased expression of the hepatic markers HNF4A, ALB, CYP3A4, and ASGR1. Differentiated organoids display upregulated albumin secretion and CYP3A4 activity relative to their undifferentiated counterparts.

The medium is supplied as HepatiCult™ Organoid Basal Medium (Human) and HepatiCult™ Organoid Differentiation Supplement (Human). The basal medium is based on DMEM/F-12 with 15 mM HEPES buffer, is stored at 2–8 °C, and is stable for 2 years from the date of manufacture. The supplement is stored at -20 °C, is stable for 2 years from the date of manufacture, and contains material derived from human plasma; donors have been tested and found negative for HIV-1, HIV-2, hepatitis B, and hepatitis C, but the product should be treated as potentially infectious and handled in accordance with universal precautions.

This product was developed under license to intellectual property owned by Hubrecht Organoid Technology (HUB) and is sold for research use only. Use of the product for growing organoids for drug screening with the aim of commercial gain, or for other commercial purposes, requires a separate Organoid-Growth License from HUB.
