CR Medium, 250 mL

STEMCELL Technologies CR Medium is a specialized cell culture medium designed for the conditional reprogramming (CR) of epithelial cells. In CR culture, epithelial cells are seeded onto an irradiated 3T3-J2 mouse fibroblast feeder layer in the medium, which contains Y-27632, an inhibitor of Rho-associated coiled-coil containing protein kinases ROCK1 and ROCK2. Under these conditions, epithelial cells are reprogrammed into a highly proliferative, undifferentiated adult epithelial stem cell state known as conditionally reprogrammed cells (CRCs), forming discrete epithelial clusters that are easily distinguished among the feeder cells.

The medium supports all types of epithelia, including simple, pseudostratified, and stratified, and the technology has been demonstrated across multiple species, including human, mouse, rat, pig, horse, rabbit, dog, and fish. CR Medium is used together with 3T3-J2 Irradiated Feeder Cells to support epithelial cell expansion, and can also serve as a base for generating conditioned medium for in vitro expansion of primary epithelial cells. Applications enabled by this technology include biobanking, disease modeling, drug discovery and development, and regenerative medicine.
