CloneR2

This defined, serum-free TeSR™ supplement from STEMCELL Technologies, known as CloneR™2, is designed to increase the cloning efficiency and single-cell survival of human embryonic stem (ES) and induced pluripotent stem (iPS) cells. It supports the generation of clonal ES and iPS cell lines whether used with low-density seeding (< 25 cells/cm²) or FACS-based single-cell sorting at 1 cell/well, and it also improves the post-thaw recovery of cryopreserved cells.

Across four hPSC cell lines, CloneR™2 delivered an average cloning efficiency of 38.6 ± 6.0% compared with 21.2 ± 5.6% for the original CloneR™ supplement. When used as a seeding supplement for the first 24 hours of culture, CloneR™2 produced an average seeding efficiency of 98.2 ± 12.8% versus 81.9 ± 15.8% with Y-27632. In single-cell passaging experiments, CloneR™2 yielded an average fold-expansion of 49.1 ± 10.4 compared with 21.1 ± 8.2 for Y-27632.

CloneR™2 is intended for research use only. It should be stored at -20°C and is stable for 1 year from the date of manufacture. The supplement is designed for use with mTeSR™1, mTeSR™ Plus, TeSR™-AOF, or TeSR™-E8™ media.
