CloneR, 5 Pack

A defined, animal component-free supplement from STEMCELL Technologies, CloneR is engineered to support single-cell cloning workflows for human pluripotent stem cells (hESCs and hiPSCs).

It is added to cell culture medium immediately after cell dissociation and through the early expansion phase, replacing methods that rely on ROCK inhibitors or mouse embryonic fibroblast (MEF) feeders. CloneR improves attachment, survival, and proliferation of dissociated single hPSCs, making it particularly useful for clonal expansion following gene editing steps such as CRISPR-Cas9, CRISPR-Cas12a, or TALEN-mediated modifications.

Each 5 Pack includes five single-use vials intended for long-term freezer storage at -20 °C, with stability preserved through repeated use within the recommended window once thawed. The formulation reduces spontaneous differentiation and minimizes apoptosis triggered by dissociation stress, supporting more uniform colonies during the critical 7–14 day post-plating interval.

Typical use: dilute CloneR 1:100 in mTeSR Plus or a similar hPSC maintenance medium, apply to freshly plated single cells, and replace with maintenance medium without CloneR once colonies reach approximately 100–200 µm in diameter. CloneR is compatible with Matrigel, vitronectin, and other defined matrices used in feeder-free culture.
