Methylcellulose-based semi-solid medium from STEMCELL Technologies streamlines hybridoma generation by consolidating HAT selection and single-cell cloning into a single plating step after PEG-mediated fusion of myeloma partners with primed B-lymphocytes. Fused cells are simply resuspended in the viscous matrix and dispensed into Petri dishes; discrete hybridoma colonies grow embedded within the medium and are typically ready for manual or robotic picking within 10 to 14 days when cultured at 37 degrees C with 5% CO2.

Each bottle delivers 90 mL of ready-to-use complete medium with hypoxanthine, aminopterin, and thymidine selective agents already incorporated, eliminating the need for separate limiting dilution or soft agar overlay procedures downstream of fusion. The medium should be stored frozen at -20 degrees C and protected from light, with thawed aliquots kept refrigerated to preserve colony-forming performance across monoclonal antibody discovery workflows.

Every lot is functionally tested for cloning efficiency against validated myeloma fusion partners, supporting reproducible hybridoma recovery from cell fusion through clonal isolation. The reagent integrates naturally with other STEMCELL Technologies hybridoma products, including PEG fusion agent and compatible myeloma lines, to support end-to-end antibody development.
