This small molecule from STEMCELL Technologies is a Janus kinase 3 (JAK3) inhibitor with reported effects on cancer cell survival, macrophage stimulation, and neural stem cell (NSC) growth. The compound is also known by the synonym Janex-1 and has the chemical name 4-[(6,7-dimethoxy-4-quinazolinyl)amino]-phenol. It is a crystalline solid with the molecular formula C16H15N3O3 and a molecular weight of 297.3 g/mol.

Reported biochemical activity includes IC50 values of 9 μM against human JAK3 and 78 μM against mouse JAK3, as well as inhibition of other kinases including epidermal growth factor receptor (EGFR) in the nanomolar range, with no significant inhibition observed against JAK1 or JAK2. Approximate solubility values cited in the safety data sheet are 10 mg/ml in MeOH, 1 mg/mL in DMSO, and 1.5 mg/ml in DMF, and the product purity is reported as ≥ 98%.

Published research applications include induction of apoptosis and cell death in the human glioblastoma cell lines U373 and U87, inhibition of lipopolysaccharide (LPS)-induced nitric oxide synthase expression and nitric oxide production in macrophages, and inhibition of proliferation of neural stem cell-containing neurospheres at lower concentrations than those required to inhibit astrocyte culture proliferation. The compound is associated with the JAK/STAT signalling pathway.
