A small-molecule splicing modulator supplied as a 250 ug lyophilized powder in a single vial, UM729 inhibits the spliceosomal factor SF3B1 and is widely used to enhance somatic cell reprogramming as well as the ex vivo expansion of human hematopoietic stem and progenitor cells. In protocols distributed by STEMCELL Technologies, UM729 is typically paired with the complement modulator UM171 in defined, serum-free culture medium to support long-term maintenance of cord-blood-derived CD34+ cells and to boost induced pluripotent stem cell (iPSC) colony formation from fibroblasts and blood progenitors. The compound is reconstituted in DMSO, stored at -20 degrees C protected from light, and is intended for research use only in cell culture workflows. Working concentrations commonly fall in the 500 nM to 1 uM range during reprogramming and HSPC expansion assays, with media changes every 2 to 3 days to maintain splicing-modulator activity over a 7 to 14 day induction window.
