STEMCELL Technologies offers TWS119, a small molecule described as a WNT pathway activator that functions through inhibition of glycogen synthase kinase 3 beta (GSK3β). It is a potent disubstituted pyrrolopyrimidine inhibitor of GSK3β with an IC₅₀ of 30 nM and a Kd of 126 nM.

The compound has the chemical name 3-[[6-(3-aminophenyl)-7H-pyrrolo[2,3-d]pyrimidin-4-yl]oxy]phenol, a molecular formula of C₁₈H₁₄N₄O₂, and a molecular weight of 318.3 g/mol. It is supplied as a crystalline solid with a purity of ≥ 90%. In published applications, TWS119 has been used to maintain the bi-potent, quiescent state in hepatic stellate cells of Wistar rats, to induce differentiation of mouse embryonal carcinoma and embryonic stem cells to neurons, and to induce production of T memory stem cell-like (TSCM) cells from mouse or human CD8+ T cells, with evidence of increased persistence, proliferation, and anti-tumor activity after adoptive transfer of mouse-derived TSCM cells. The compound has also been reported to inhibit cell proliferation and induce apoptosis in human alveolar rhabdomyosarcoma cells.

The product is stable at ≤20°C as supplied and should be protected from prolonged exposure to light; it remains stable as supplied for 12 months from date of receipt. Reported solubility is ≤ 60 mM in DMSO and ≤ 0.9 mM in absolute ethanol, and the compound has low solubility in aqueous media. For cell culture use, stock solution should be diluted into culture medium immediately before use, and a final DMSO concentration above 0.1% should be avoided due to potential cell toxicity.
