Description

TWS119 from STEMCELL Technologies is a potent disubstituted pyrrolopyrimidine inhibitor of glycogen synthase kinase 3 beta (GSK3β), with an IC₅₀ of 30 nM and a Kd of 126 nM. Because GSK3 is a serine/threonine kinase that acts as a key inhibitor of the WNT pathway, TWS119 functions as a WNT pathway activator.

The compound is supplied as a crystalline solid with the chemical formula C₁₈H₁₄N₄O₂, a molecular weight of 318.3 g/mol, a purity of ≥ 90%, and CAS number 601514-19-6. It is also referenced as GSK 3B Inhibitor XII. Reported solubility is ≤ 60 mM in DMSO and ≤ 0.9 mM in absolute ethanol, and the compound has low solubility in aqueous media. As supplied, the product is stable at -20°C and should be protected from prolonged exposure to light, remaining stable for 12 months from the date of receipt.

Published applications include maintenance of the bi-potent, quiescent state in hepatic stellate cells of Wistar rats, induction of neuronal differentiation from mouse embryonal carcinoma and embryonic stem cells, and induction of T memory stem cell-like (T-SCM) cells from mouse or human CD8+ T cells, with reported increases in persistence, proliferation, and anti-tumor activity after adoptive transfer of mouse-derived T-SCM cells. In cancer research, TWS119 has been reported to inhibit cell proliferation and induce apoptosis in human alveolar rhabdomyosarcoma cells.
