An ATP-competitive small molecule inhibitor of the mammalian target of rapamycin (mTOR) supplied by STEMCELL Technologies, this compound inhibits the mTORC1 and mTORC2 complexes with IC₅₀ values of 2 nM and 10 nM, respectively. It is selective for mTOR versus a panel of over 400 other kinases screened at 10 µM.

Provided as a crystalline solid with ≥ 95% purity, the compound has a molecular weight of 607.6 g/mol and the chemical formula C₃₅H₂₈F₃N₅O₂ (CAS 1222998-36-8). For stock preparation, it can be resuspended in N-methyl-2-pyrrolidine (NMP) at concentrations up to 40 mM, though it has low solubility in aqueous media and the final NMP concentration should not exceed 0.1% in cell culture due to potential cell toxicity. The product is stable for 12 months from the date of receipt when stored at -20°C and protected from prolonged light exposure.

Published cancer research applications include inhibition of tumor growth, likely through a cytostatic rather than cytotoxic mechanism, in a U87MG PTEN-null glioblastoma mouse xenograft model, as well as hindrance of growth, motility, invasion, and survival of primary colon cancer stem-like cells in vitro and suppression of tumor growth in xenografts without affecting normal colon stem cells.
