Description
This small molecule from STEMCELL Technologies is a selective inhibitor of Rho-associated coiled-coil containing protein kinase (ROCK1 and ROCK2), a serine/threonine kinase that plays a role in cell polarity, contraction, and actin cytoskeleton reorganization. The compound, also known as Tzv, has the chemical name N-(phenylmethyl)-2-(4-pyrimidinylamino)-4-thiazolecarboxamide (CAS 1226056-71-8) and the molecular formula C15H13N5OS, with a molecular weight of 311.4 g/mol and a stated purity of ≥98%. It is supplied as a crystalline solid and is described as having low solubility in aqueous media, with reported solubilities of ~0.5 mg/mL in a 1:1 solution of DMSO:PBS (pH 7.2), ~1 mg/mL in EtOH, ~14 mg/mL in DMSO, and ~10 mg/mL in DMF. Solubility in absolute ethanol is ≤ 3.2 mM and in DMSO is ≤ 40 mM. For example, a 10 mM stock solution in DMSO can be prepared by resuspending 1 mg in 321 μL of DMSO, and stock solutions should be diluted into culture medium immediately before use, with final DMSO concentration kept at or below 0.1% to avoid potential cell toxicity. The product is reported to be effective at 5-fold lower concentrations than the related ROCK inhibitor Y-27632.
Published applications include promoting the survival of human embryonic stem (ES) cells during dissociation by stabilizing E-cadherin and improving cell attachment, and promoting the survival of single human induced pluripotent stem (iPS) cells during transfection for TALEN-mediated genome editing. The compound also increases the efficiency of reprogramming human somatic cells to iPS cells when used in combination with other small molecules, and improves reprogramming of human cord blood mononuclear cells to iPS cells. The product is stable at -20°C as supplied, should be protected from prolonged exposure to light, and is stable as supplied for 12 months from the date of receipt. Stock solutions are generally stored in DMSO at -20°C and aliquoted into working volumes to avoid repeated freeze-thaw cycles.
According to the manufacturer's safety data sheet, the compound is classified as harmful if swallowed (Acute toxicity - Oral Category 4) with an ATEmix (oral) of 500.00 mg/kg.
