Description

A small-molecule research compound supplied by STEMCELL Technologies, Thiazovivin is described as a selective inhibitor of Rho-associated coiled-coil containing protein kinase (ROCK), a serine/threonine kinase that plays a role in cell polarity, contraction, and actin cytoskeleton reorganization. The product is reported to be effective at 5-fold lower concentrations than another common ROCK inhibitor, Y-27632.

The chemical name is N-benzyl-2-(pyrimidin-4-ylamino)-1,3-thiazole-4-carboxamide (synonym: N-(phenylmethyl)-2-(4-pyrimidinylamino)-4-thiazolecarboxamide), with CAS Number 1226056-71-8, molecular formula C15H13N5OS, and molecular weight 311.4 g/mol. The material is supplied as a crystalline solid with a stated purity of ≥ 98%. Reported solubilities include ~0.5 mg/mL in a 1:1 solution of DMSO:PBS (pH 7.2), ~1 mg/mL in EtOH, ~14 mg/mL in DMSO, and ~10 mg/mL in DMF.

Published applications include promoting survival of human embryonic stem cells during dissociation by stabilizing E-cadherin and improving cell attachment, promoting survival of single human induced pluripotent stem cells during transfection for TALEN-mediated genome editing, and increasing the efficiency of reprogramming human somatic cells and cord blood mononuclear cells to iPS cells. The product is stable at -20 °C as supplied, should be protected from prolonged exposure to light, and is stable as supplied for 12 months from date of receipt. Stock solutions are recommended at ≤ 40 mM in DMSO or ≤ 3.2 mM in absolute ethanol. The compound is classified as harmful if swallowed and is intended for research use only.
