Description

From STEMCELL Technologies, this small molecule is supplied as a crystalline solid and is a sesquiterpene lactone that acts as a cell-permeable inhibitor of sarco-endoplasmic reticulum Ca2+-ATPases (SERCAs). The 5 mg format has a stated purity of ≥ 97%, a chemical formula of C34H50O12, a molecular weight of 650.8 g/mol, and CAS Number 67526-95-8.

In published research, the compound has been used to induce autophagy and endoplasmic reticulum stress in mammalian cells, and to induce apoptosis in rat thymocytes. The product is stable for 12 months from the date of receipt when stored at -20°C and protected from prolonged exposure to light; for long-term storage, a desiccant is recommended. Reported solubility is ≤ 45 mM in DMSO and ≤ 45 mM in absolute ethanol. For example, a 10 mM stock solution in DMSO is prepared by resuspending 1 mg in 154 μL of DMSO. Stocks should be freshly prepared, stored at -20°C in DMSO, and aliquoted to avoid repeated freeze-thaw cycles. The compound has low solubility in aqueous media.

For use as a cell culture supplement, the stock should be diluted into culture medium immediately before use, keeping the final DMSO concentration below 0.1% to avoid potential cell toxicity. According to the safety data sheet, Thapsigargin is classified as hazardous: it causes skin irritation (Category 2), serious eye irritation (Category 2A), respiratory sensitization (Category 1), and may cause respiratory irritation from single exposure (Category 3). It should be handled with protective gloves, eye protection, and in a well-ventilated area. For research use only.
