STEMvision Mouse M3434 Assay

An image-analysis package from STEMCELL Technologies, the STEMvision Mouse M3434 Assay is designed to count total hematopoietic colonies—those produced by combined myeloid and erythroid progenitors—in mouse bone marrow cultures plated in MethoCult™ GF M3434 medium. It runs on the STEMvision™ system, which combines dedicated imaging hardware with software-based colony scoring and replaces manual counting under an inverted microscope. Standardized algorithms help reduce operator-to-operator variability when scoring colony-forming unit (CFU) assays.

The Mouse M3434 Assay is one of three mouse analysis packages offered for the instrument; companion packages score myeloid-only progenitors in M3534 and erythroid-only progenitors in M3436. It works with meniscus-free SmartDish™ cultureware for more accurate counting of hematopoietic colonies in CFU assays. In the manufacturer's validation work, automated counts using this package have been described as highly correlated with manual inverted-microscope counts of mouse BM CFU assays, with culture reading recommended between days 10 and 12. The Mouse M3434 Assay is sold separately and requires the STEMvision™ instrument.
