Annexin V, FI,100tests

STEMCELL Technologies offers an apoptosis staining and detection kit that combines Annexin V conjugated to FITC with 7-AAD viability dye and Annexin V Binding Buffer for use in flow cytometry. Annexin V belongs to the annexin family of intracellular proteins that bind membrane phospholipids in a calcium-dependent manner and show high affinity for phosphatidylserine (PS), which resides in the inner leaflet of the plasma membrane. During early-stage apoptosis, PS is translocated from the inner to the outer leaflet of the cell membrane, exposing it to the external cellular environment where it can be detected by Annexin V; this event precedes any loss of membrane integrity. As cells progress toward late-stage apoptosis or necrosis, the plasma membrane becomes compromised, permitting the 7-AAD viability dye to enter the cell. Early apoptotic cells therefore stain positive for Annexin V and negative for the viability dye, whereas late apoptotic or necrotic cells stain positive for both markers.

The kit is verified and reported for flow cytometry (FC) applications. Representative data were generated by flow cytometric analysis of C57BL/6 mouse thymocytes incubated with dexamethasone overnight and subsequently labeled with FITC-conjugated Annexin V and 7-AAD. All components are intended for research use only and are not for human or animal diagnostic or therapeutic applications.

All components should be stored at 2 - 8°C, protected from prolonged exposure to light, and not frozen; each component remains stable until the expiry date printed on its label.
