Description

STEMCELL Technologies offers an apoptosis detection kit that enables combined identification of early- and late-stage apoptotic cells by flow cytometry. The complete kit includes Annexin V conjugated to APC, the 7-Aminoactinomycin D (7-AAD) viability dye, and Annexin V Binding Buffer, supplied together as a single set.

Annexin V is a member of the annexin family of intracellular proteins that bind membrane phospholipids in a calcium-dependent manner. The reagent has a high affinity for phosphatidylserine (PS), which is normally located in the inner leaflet of the plasma membrane. During early-stage apoptosis, PS is translocated from the inner to the outer leaflet of the cell membrane, exposing it to the external environment where Annexin V can detect it. This PS translocation occurs prior to loss of membrane integrity. As cells progress through apoptosis and toward necrosis, the cell membrane becomes compromised and the 7-AAD viability dye passes into the cell. Cells undergoing early apoptosis therefore stain positive for Annexin V and negative for the viability dye, whereas late apoptotic or necrotic cells stain positive for both Annexin V and 7-AAD.

The kit has been verified for flow cytometry applications. Representative data were generated using C57BL/6 mouse thymocytes incubated at 37°C with 1 µM dexamethasone overnight, then harvested and labelled with Annexin V, APC and 7-AAD. All components should be stored at 2 - 8°C, protected from prolonged exposure to light, and not frozen; they remain stable until the expiration date printed on the label. The product is intended for research use only.
