Description

A small molecule from STEMCELL Technologies, StemRegenin 1 (SR1) is an antagonist of the aryl hydrocarbon receptor (AHR) that promotes ex vivo expansion of CD34+ human hematopoietic stem cells and supports their maintenance in culture. The compound is supplied as a crystalline hydrochloride salt (C₂₄H₂₃N₅OS · HCl, CAS 2319882-01-2) with a molecular weight of 466.0 g/mol and a purity of ≥98%.

Designed as a cell culture supplement, it is applied in serum-free medium for the culture and expansion of hematopoietic cells. Published research uses include promoting maintenance and self-renewal of human hematopoietic stem cells, stimulating differentiation of CD34+ hematopoietic progenitor cells into functional human dendritic cells, supporting hematopoietic differentiation of induced pluripotent stem (iPS) cells, and collaborating with UM729 to prevent differentiation of acute myeloid leukemia (AML) cells in culture.

The compound is soluble in DMSO at concentrations ≤10 mM; for instance, a 5 mM stock solution can be prepared by dissolving at 429 μL of DMSO per mg. Stock solutions should be freshly prepared, stored at -20°C, protected from light, and aliquoted to avoid repeated freeze-thaw cycles. It has low solubility in aqueous media, though concentrations up to 1 µM can be reached by dilution from a DMSO stock. Final DMSO concentration in culture should not exceed 0.1%. The product is stable as supplied at -20°C for 12 months from date of receipt when protected from prolonged exposure to light.
