Description

Provided by STEMCELL Technologies, this small molecule is a bromo-substituted quinazoline first identified as a small-molecule enhancer of rapamycin (SMER). It induces autophagy by increasing autophagosome synthesis and enhancing the clearance of model autophagy substrates.

The chemical name is 6-bromo-N-prop-2-enylquinazolin-4-amine, with a molecular formula of C₁₁H₁₀BrN₃ and a molecular weight of 264.1 g/mol (CAS Number 307538-42-7). The product is supplied as a crystalline solid with a stated purity of ≥ 98%. It is stable at -20°C as supplied and should be protected from prolonged exposure to light; it remains stable as supplied for 12 months from the date of receipt. Solubility information indicates DMSO ≤ 110 mM and absolute ethanol ≤ 110 mM, and the compound has low solubility in aqueous media.

Published applications include use as a component of a neural reprogramming cocktail that reprograms mouse fibroblasts into induced neural stem cell-like cells. In disease modeling, the compound enhances clearance of autophagy substrates including mutant huntingtin in Huntington's disease and A53T α-synuclein in familial Parkinson's disease. Through autophagy factor ATG5, it stimulates erythropoiesis and up-regulates expression of globin genes in in vitro and in vivo models of Diamond-Blackfan anemia, and decreases levels of Alzheimer's disease indicators (amyloid-β peptide and the APP-derived fragment) via the autophagy-related protein 5-dependent pathway. The manufacturer also describes this product as a PI3K/AKT pathway inhibitor (inhibits AKT) and an Activin/Nodal/TGFβ pathway inhibitor (inhibits ALK2). Products are for research use only.
