Description

From STEMCELL Technologies, this small molecule is a WNT pathway activator and a cell-permeable, ATP-competitive inhibitor of glycogen synthase kinase 3 (GSK3), with activity reported against both the GSK3α (IC₅₀ = 34 nM) and GSK3β isozymes. The chemical identity is 3-(2,4-Dichlorophenyl)-4-(1-methyl-1H-indol-3-yl)-1H-pyrrole-2,5-dione (CAS 280744-09-4), corresponding to the molecular formula C₁₉H₁₂Cl₂N₂O₂ and a molecular weight of 371.2 g/mol. The material is supplied as a crystalline solid with a stated purity of ≥ 98%.

The product is stable at -20 °C as supplied and should be protected from prolonged exposure to light. Reported solubility in DMSO is ≤ 5.3 mM, and the compound has low solubility in aqueous media. For general guidance, stock solutions are recommended to be prepared fresh in DMSO at -20 °C, aliquoted to limit freeze-thaw cycles, with stability assessed per individual application. When added to cell culture, it should be introduced immediately before use, keeping the final DMSO concentration below 0.1% to minimise potential cell toxicity.

Published research applications span maintenance and self-renewal of stem cells, including maintenance of mouse embryonic stem cells in an undifferentiated state without LIF, expansion of mouse retinal stem cells, increased neural progenitor proliferation, modulation of neural stem cell division symmetry, inhibition of adipocyte differentiation in mesenchymal stem cells, and promotion of haematopoietic stem cell development in AGM explants. Reported differentiation uses include enhancement of insulin-induced myoblast differentiation, stimulation of neural stem cell differentiation, induction of neuronal differentiation in cultured human neural progenitor cells, and promotion of dendritic cell differentiation from haematopoietic progenitors. In cancer research, the compound has been used to induce differentiation and reduce the cancer stem cell population in cultured human glioblastoma cells.
