A selective, potent, cell-permeable inhibitor of p38 MAP kinases from STEMCELL Technologies, supplied as a crystalline solid with ≥98% purity. The compound inhibits p38α (SAPK2A, MAPK14) and p38β (SAPK2B, MAPK11) with IC50 values of 50 and 100 nM, respectively. When tested at 10 µM, it has negligible effects on a range of other kinases, including ERKs and JNKs. As a pyridinyl imidazole, it directly binds p38 MAP kinases in the ATP binding pocket.

Published research applications include improving the self-renewal ability of neural stem cells from NPC1-deficient mice, blocking adiponectin-mediated proliferation of hematopoietic stem cells, reducing BMP3-mediated proliferation of C3H10T1/2 mesenchymal stem cells, and inducing cardiomyocyte differentiation from human embryonic stem cells.

The compound has low solubility in aqueous media. It is soluble in absolute ethanol (≤750 µM) and DMSO (≤45 mM). For cell culture use, stock solution should be diluted into culture medium immediately before use, with final DMSO concentration kept below 0.1% to avoid potential cell toxicity. Store at -20°C and protect from prolonged exposure to light.
