Description

STEMCELL Technologies supplies this cell-permeable small molecule as a direct antagonist of Smoothened (SMO) and an inhibitor of the Sonic hedgehog (Shh) signaling pathway. It binds Smoothened with a reported Ki of 2.4 nM (Chen et al.; Rominger et al.). The compound is provided as a crystalline solid with the chemical name N-[(3,5-dimethyl-1-phenyl-1H-pyrazol-4-yl)methylene]-4-(phenylmethyl)-1-piperazinamine, CAS number 304909-07-7, molecular formula C23H27N5, and molecular weight 373.5 g/mol. Reported purity is ≥ 98%.

Solubility is stated as ≤ 8 mM in DMSO and ≤ 13 mM in absolute ethanol, with low solubility in aqueous media. For cell culture use, stock solution should be diluted into culture medium immediately before use, with final DMSO concentration kept below 0.1% to avoid potential cell toxicity. The product is stable at -20°C as supplied, protected from prolonged exposure to light, and for long-term storage should be kept with a desiccant; it is reported stable for 12 months from date of receipt when stored appropriately. According to the manufacturer's Safety Data Sheet, the substance is not classified as hazardous under GHS or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200).

Published research applications include promoting beta cell differentiation from human embryonic stem cells (Yung et al.) and blocking hedgehog signaling to inhibit tumorigenesis and proliferation in non-small-cell lung cancer cells (Bai et al.). The product is intended for research use only.
