Description

From STEMCELL Technologies, this small molecule is a cell-permeable, selective inhibitor of Type 4 cyclic nucleotide phosphodiesterases (PDE4), which mediate cyclic AMP (cAMP) degradation. It preferably inhibits the PDE4A isoform with an IC₅₀ of 3 nM, compared to 130 nM for PDE4B and 240 nM for PDE4D, and inhibits interferon (IFN)-γ-stimulated phosphorylation of p38 mitogen-activated protein (MAP) kinase through PDE4B and/or PDE4D inhibition.

The compound, also referenced as ZK 62711 and SB 95952, is identified by CAS number 61413-54-5 and chemical formula C₁₆H₂₁NO₃, with a molecular weight of 275.3 g/mol. It is supplied as a crystalline solid at a purity of ≥ 98%. The product is stable at -20 °C as supplied and should be protected from prolonged exposure to light. Solubility data indicate ≤ 35 mM in DMSO and ≤ 15 mM in absolute ethanol, with low solubility in aqueous media. For use as a cell culture supplement, stock solution should be diluted into culture medium immediately before use, with final DMSO concentration kept below 0.1% due to potential cell toxicity.

Published research applications include differentiation (enhancing BMP-2-induced osteoblastic differentiation of mouse mesenchymal stem cells and inducing neural differentiation of human bone marrow-derived mesenchymal stem cells), reprogramming (inducing reprogramming of adult human dermal fibroblasts into expandable induced neuronal stem cells in combination with A83-01, CHIR99021, sodium butyrate, LPA, SP600125, and exogenous OCT4 expression), disease modeling (promoting survival of newly formed mouse hippocampal neurons in a model of ischemia and reversing amphetamine-induced reductions in auditory-evoked potentials in a C57BL/6J mouse model of schizophrenia), and immunology (suppressing C5a-stimulated leukotriene C4 synthesis in human eosinophils and inhibiting lipopolysaccharide-induced tumor necrosis factor synthesis in human monocytes).
