A small molecule inhibitor from STEMCELL Technologies, R428 is a potent and selective inhibitor of the receptor tyrosine kinase Axl with an IC50 of 14 nM and antiproliferative activity. Axl is involved in cell survival, proliferation, adhesion, and migration. The compound exhibits over 100-fold selectivity for Axl over the insulin receptor, epidermal growth factor receptor, human epidermal growth factor receptor 2, and platelet-derived growth factor receptor β.

Also known as Bemcentinib or BGB 324, it carries the CAS number 1037624-75-1, a chemical formula of C30H34N8, and a molecular weight of 506.6 g/mol, with a reported purity of ≥ 98%. It is supplied as a crystalline solid and remains stable at -20°C for 12 months from the date of receipt when protected from prolonged light exposure and stored with a desiccant for long-term use.

The compound is soluble in DMSO up to 9.8 mM but has low solubility in aqueous media. Stock solutions should be freshly prepared; when used as a cell culture supplement, the stock should be diluted into culture medium immediately before use, and the final DMSO concentration should not exceed 0.1% due to potential cell toxicity.
