Description

A selective, cell-permeable inhibitor of the MEK/ERK pathway supplied by STEMCELL Technologies. The compound is 2-(2-amino-3-methoxyphenyl)-4H-1-benzopyran-4-one (CAS 167869-21-8; molecular formula C16H13NO3; molecular weight 267.3 g/mol) with a stated purity of ≥98%, supplied as a crystalline solid. It prevents the activation of MEK1 (IC50 = 2 - 7 µM) and MEK2 (IC50 = 50 µM) by upstream kinases and does not inhibit activated MEK or the p38 MAPK pathway.

According to the supplier, the product is stable for 12 months from the date of receipt when stored at -20°C, and should be protected from prolonged exposure to light. It is soluble in DMSO at concentrations up to 70 mM; stock solutions are typically prepared in fresh DMSO, for example a 10 mM stock by resuspending 1 mg in 374 µL of DMSO. The supplier notes low solubility in aqueous media and recommends diluting stock solution into culture medium immediately before use, keeping the final solvent concentration at or below 0.1% to avoid potential cell toxicity.

The Safety Data Sheet classifies the substance as acutely toxic if swallowed (oral ATEmix 100.00 mg/kg) and advises storing the material locked up and out of the reach of children. Published research applications highlighted by the supplier include enhancing the growth and self-renewal of mouse embryonic stem cells, enhancing the adipogenic differentiation of human mesenchymal stem cells while blocking their osteogenic differentiation, and decreasing the number of AML blast colonies with minimal effect on normal hematopoietic progenitors. For research use only.
