Description

A selective and potent ATP-competitive inhibitor of fibroblast growth factor receptor (FGFR) is offered by STEMCELL Technologies. It acts on both FGFR3 and FGFR1 with IC50 values of 5 nM and 21.5 nM, respectively, and also inhibits FGFR2, FGFR4, and vascular endothelial growth factor receptor 2 (VEGFR2). The compound is approximately 1000 times more potent than the FGFR inhibitor SU5402, while showing little to no activity against PDGFR, EGFR, MEK, or PKC.

The supplied material is a crystalline solid with the chemical formula C28H41N7O3, a molecular weight of 523.7 g/mol, and the chemical name N-[2-[[4-(diethylamino)butyl]amino]-6-(3,5-dimethoxyphenyl)pyrido[2,3-d]pyrimidin-7-yl]-N'-(1,1-dimethylethyl)-urea (CAS 219580-11-7). Reported purity is ≥ 95%. For storage, the product is stable at -20°C as supplied and should be protected from prolonged exposure to light, remaining stable for 12 months from the date of receipt. The compound is soluble in DMSO at concentrations up to 20 mM and exhibits low solubility in aqueous media. For cell culture applications, stock solutions should be freshly prepared in DMSO and diluted into culture medium immediately before use, with final DMSO concentration kept below 0.1% to avoid potential cell toxicity.

In published research, PD173074 has been used to suppress differentiation and maintain the undifferentiated state of mouse embryonic stem cells, to prevent excision-mediated differentiation in mouse induced pluripotent stem cells, and—in combination with OCT4, KLF4, KLF2, LIF, CHIR99021, and PD0325901—to promote reprogramming of human ES cells to a naïve state. The compound also blocks neural differentiation of mouse ES cells and can promote differentiation of human ES cells that are not in a naïve or ground state.
