Description

A selective, cell-permeable inhibitor of the MEK/ERK pathway supplied by STEMCELL Technologies, this small molecule blocks the activation and downstream signaling of MEK. It is described as an extremely potent inhibitor that suppresses phosphorylation of ERK in C26 cells at very low concentrations, with an IC50 of 0.33 nM.

The compound is provided as a crystalline solid with a purity of ≥ 98% and a molecular weight of 482.2 g/mol. Its CAS number is 391210-10-9, and the chemical name is N-[(2R)-2,3-dihydroxypropoxy]-3,4-difluoro-2-[(2-fluoro-4-iodophenyl)amino]-benzamide. The compound is soluble in DMSO at ≤ 20 mM; to prepare a 10 mM stock solution, resuspend 1 mg in 207 µL of fresh DMSO. It should be stored at -20°C, protected from prolonged exposure to light, and is stable as supplied for 12 months from receipt. Stock solutions should be freshly prepared, aliquoted to limit freeze-thaw cycles, and the final DMSO concentration in cell culture should be kept below 0.1% to avoid toxicity.

Published research applications include maintaining undifferentiated mouse embryonic stem (ES) cells in combination with CHIR99021 in the absence of LIF, supporting the derivation and maintenance of rat ES cells, selecting and expanding fully reprogrammed mouse induced pluripotent stem (iPS) cells at later stages of reprogramming, increasing the efficiency of reprogramming human somatic cells to iPS cells in combination with SB431542 and Thiazovivin, promoting reprogramming of human somatic cells using OCT4 alone, and generating ground-state iPS cells from human and rat somatic cells alongside CHIR99021 and A 83-01. The product is intended for research use only and is classified as hazardous.
