Description

Available from STEMCELL Technologies, NU7026 is a small molecule inhibitor of DNA-dependent protein kinase (DNA-PK), an enzyme that plays a key role in the non-homologous end joining (NHEJ) DNA repair pathway. The compound is supplied as a crystalline solid with a purity of ≥ 95% and is identified by CAS Number 154447-35-5. It has a chemical formula of C₁₇H₁₅NO₃, a molecular weight of 281.3 g/mol, and is also known as LY293646 or DNA-PK Inhibitor II.

This inhibitor has demonstrated utility in genome editing applications, where it increases the efficiency of homology-directed repair (HDR) at the expense of NHEJ in human pluripotent stem cells, supporting CRISPR-Cas9 workflows. In cancer research, the compound sensitizes human cancer cell lines to DNA double-strand break-inducing therapies, including chemo- and radio-therapy, by inhibiting DNA-PK activity and inducing cell cycle arrest at the G2/M phase. Earlier work also showed that NU7026 sensitizes cells to radiation, suggesting potential for anticancer therapies.

The product is stable at -20°C as supplied and should be protected from prolonged exposure to light. For long-term storage, the compound should be kept with a desiccant and remains stable as supplied for 12 months from the date of receipt. NU7026 has low solubility in aqueous media; in DMF it is soluble at ≤ 530 µM. For use as a cell culture supplement, stock solutions should be diluted into culture medium immediately before use, avoiding final DMF concentrations above 0.1% due to potential cell toxicity.
