Description

This small molecule from STEMCELL Technologies is a potent inhibitor of DNA-dependent protein kinase (DNA-PK), an enzyme that plays a central role in the non-homologous end joining (NHEJ) DNA repair pathway. Supplied as a crystalline solid, the chemical name is 2-(4-morpholinyl)-4H-naphtho[1,2-b]pyran-4-one with a molecular weight of 281.3 g/mol and molecular formula C17H15NO3. It is also known by the synonyms LY293646 and DNA-PK Inhibitor II (CAS 154447-35-5) and is supplied at a purity of ≥ 95%.

In research applications, the compound sensitizes cells to radiation and has potential for anticancer therapies. It increases the efficiency of homology-directed repair (HDR) in CRISPR-Cas9 genome editing, promoting precise genome editing in human pluripotent stem cells by shifting repair away from NHEJ. In cancer research, it sensitizes human cancer cell lines to DNA double-strand break-inducing therapy (chemo- or radio-therapy) by inhibiting DNA-PK activity and inducing cell cycle arrest at G2/M phase.

The product is stable at -20°C as supplied and should be protected from prolonged exposure to light. For long-term storage, it should be kept with a desiccant, where it remains stable for 12 months from the date of receipt. Solubility in DMF is 0.15 mg/mL, and a stock solution guide suggests ≤ 530 µM in DMF. The compound has low solubility in aqueous media; for use as a cell culture supplement, the stock solution should be diluted into culture medium immediately before use, avoiding final DMF concentrations above 0.1% due to potential cell toxicity. For research use only.
