Description

From STEMCELL Technologies, this small-molecule WNT pathway inhibitor potently blocks WNT signaling and prevents WNT-induced accumulation of β-catenin by stabilizing AXIN2, a member of the β-catenin destruction complex. In a cell-based WNT/β-catenin pathway reporter assay, the compound inhibits the WNT response with an IC₅₀ of 180 nM. The IWR-1-exo diastereomer exhibits much less activity against the WNT/β-catenin pathway and has been used as a control.

Supplied as a crystalline solid with a purity of ≥ 98%, IWR-1-endo has a molecular weight of 409.4 g/mol and a molecular formula of C25H19N3O3 (CAS 1127442-82-3). It is soluble at approximately 0.25 mg/mL in a 1:3 solution of DMSO:PBS (pH 7.2) and at approximately 20 mg/mL in DMF or DMSO. The product is stable for 12 months from the date of receipt when stored at -20°C and protected from prolonged exposure to light. It is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS).

Published applications include promoting self-renewal and maintaining pluripotency of human embryonic stem cells and mouse epiblast stem cells when used in combination with CHIR99021, promoting differentiation of cardiomyocytes from human pluripotent stem cells induced to mesoderm with Activin A and/or BMP4, and inducing differentiation of human PSC-derived alveolar epithelial type II (AETII) cells to AETI cells. The product is for research use only and is not intended for human or animal diagnostic or therapeutic uses.
