ISCK03, 10 mg

From STEMCELL Technologies, ISCK03 is a cell-permeable phenyl-imidazolosulfonamide compound that inhibits KIT activity. It blocks SCF-induced KIT phosphorylation and downstream ERK phosphorylation at concentrations of 1–5 µM in 501mel melanoma cells (Na et al., 2007). The product is supplied as a crystalline solid with a purity of ≥ 95% and a molecular weight of 355.5 g/mol (chemical formula C₁₉H₂₁N₃O₂S).

It is soluble in DMSO at concentrations up to 70 mM and in absolute ethanol at concentrations up to 5.6 mM, but shows low solubility in aqueous media. For use as a cell culture supplement, the stock solution should be diluted into culture medium immediately before use, and the final DMSO concentration should be kept at or below 0.1% to avoid potential cell toxicity. The compound is stable at ≤20 °C as supplied, should be protected from prolonged exposure to light, and stored with a desiccant for long-term storage.

Published applications include a significant decrease in blood vessel density in human lung adenocarcinoma (A549) cells irradiated with X-rays at 6 Gy (Kamlah et al., 2011) and reduced cell viability and ERK1/2 phosphorylation in acute myeloid leukemia (AML)-derived cell lines (Kon et al., 2013).
