Description

A cell-permeable small molecule supplied by STEMCELL Technologies, Gö6983 inhibits several isoforms of protein kinase C (PKC). Reported IC₅₀ values are 7, 7, 6, 10, 60, and 20,000 nM for PKC α, PKC β, PKC γ, PKC δ, PKC ζ, and PKC µ, respectively (Gschwendt et al.). The compound is supplied as a crystalline solid with a chemical formula of C₂₆H₂₆N₄O₃ and a molecular weight of 442.5 g/mol, and has a reported purity of ≥ 98%.

It is soluble in DMSO at concentrations up to 20 mM and is reported as stable for 12 months from the date of receipt when protected from prolonged exposure to light. For use as a cell culture supplement, the stock solution should be diluted into culture medium immediately before use, with a final DMSO concentration kept below 0.1% to avoid potential cell toxicity.

In published stem cell research, this PKC inhibitor has been used to enhance reprogramming of mouse embryonic fibroblasts to induced pluripotent stem cells (Dutta et al.) and to support direct lineage reprogramming of fibroblasts to mature neurons when combined with CHIR99021, RepSox, Forskolin, SP600125, Valproic Acid, and Y-27632 (Hu et al.). It has also been reported to inhibit differentiation and maintain pluripotency in mouse embryonic stem cells (Dutta et al.), enhance viability and growth of human ground state naive pluripotent stem cells (Gafni et al.), and inhibit proplatelet formation from megakaryocytes derived from adult mouse bone marrow (Williams et al.).
