Frozen Cord Blood PT Program - 1st, Spri

Designed as a quality control kit for hematopoietic colony assays, this proficiency testing program from STEMCELL Technologies enables laboratories to assess their performance in cord blood colony-forming unit (CFU) assays, focusing on sample preparation, assay setup, and colony assessment.

The kit includes frozen human cord blood mononuclear cells, MethoCult™ GF medium, Iscove's MDM with 2% FBS, a dry goods kit, and a session-specific information letter. Participants thaw the cell stock, perform nucleated and viable cell counts, and dilute the cells to a 10X plating density in Iscove's MDM with 2% FBS before inoculating into MethoCult™. The final plating density is dispensed into culture dishes, which are incubated at 37°C with 5% CO₂ and ≥ 95% humidity for 14 days. Colonies are then enumerated, with participants identifying BFU-E, CFU-GM, and CFU-GEMM colony types.

Data is submitted online using the program's data submission forms, and a session-specific letter provides the 10X plating density value and other details for the cohort.
