Frozen Cord Blood PT Program - 1st, Fall

A quality control kit from STEMCELL Technologies designed for use with the hematopoietic colony forming unit (CFU) assay. The program assesses participant proficiency at performing cord blood colony assays, focusing on sample preparation, assay setup, and assessment.

Kit components include Human Cord Blood Mononuclear Cells (Frozen), MethoCult™ GF medium, Iscove’s MDM with 2% FBS, a Dry Goods Kit, and a session-specific information letter. The frozen mononuclear cells must be stored at -135°C or colder; MethoCult™ GF and Iscove’s MDM with 2% FBS are stored at -20°C, and the dry goods kit is held at room temperature (15 – 25°C).

On receipt, cells are thawed in a 37°C water bath for approximately 2 minutes, washed in Iscove’s MDM with 2% FBS, and centrifuged at 300 x g for 10 minutes at room temperature (15 – 25°C). A 10X Plating Density is prepared and combined with the 5 mL tube of MethoCult™, then 1.1 mL is plated into each of 4 x 35 mm culture dishes. Cultures are incubated at 37°C, 5% CO₂, and ≥ 95% humidity for 14 days, after which colonies are enumerated. Reported colony types include BFU-E, CFU-GM, and CFU-GEMM. Data can be submitted online, by email, or via the included data submission worksheet. The product is for research use only.
