DMH1, 1 mg

A selective small-molecule inhibitor from STEMCELL Technologies, this dorsomorphin homolog 1 (DMH1) targets activin receptor-like kinase 2 (ALK2) with an IC50 of 13–108 nM, a type I bone morphogenetic protein (BMP) receptor. The compound shows no detectable inhibition of ALK4, ALK5, AMPK, KDR (VEGFR2), or PDGFRβ, while inhibiting ALK1 and ALK3 at nanomolar concentrations. It is supplied as a crystalline solid with a chemical formula of C24H20N4O, a molecular weight of 380.4 g/mol, and a purity of ≥98% (CAS 1206711-16-1). Solubility has been reported in DMF at 20 mg/mL and DMSO at 1 mg/mL, with stock solutions recommended in DMF at ≤50 mM or DMSO at ≤2.6 mM.

DMH1 promotes differentiation of pluripotent stem cells. Published applications include induction of mouse embryonic stem cells to cardiomyocyte progenitor cells, differentiation of human induced pluripotent stem cells to SOX1- and PAX6-expressing neural precursor cells, and dorsalization of the embryonic axis in early zebrafish embryos without disrupting angiogenesis. In cancer research, the compound has been reported to suppress non-small cell lung cancer cell growth, migration, and invasion in vitro and to attenuate xenografted lung tumor growth in vivo, as well as to inhibit chemotherapeutic drug-induced autophagy.
