Description
A γ-secretase inhibitor from STEMCELL Technologies, DAPT indirectly blocks Notch signaling and also influences other γ-secretase substrates including amyloid precursor protein, E-cadherin, and ErbB4. The compound is a crystalline solid with a reported purity of ≥ 95%, a molecular weight of 432.5 g/mol, and molecular formula C23H26F2N2O4 (CAS 208255-80-5). It is also referred to by the alternative names GSI-IX and LY-374973.
According to published applications, DAPT has been used to reduce colony-forming efficiency of mouse neural stem cells, enhance radiation-induced cell death of glioma stem cells, and promote the differentiation of nociceptors from human pluripotent stem cells, neurons from human and mouse embryonic stem cells, retinal pigment epithelium from mouse ES cells, and pancreatic endocrine cells from human pluripotent stem cells. In cancer research, it has been reported to reduce mammosphere-forming efficiency of breast cancer cell lines and ductal carcinoma in situ cells. The product is described as stable at -20°C as supplied when protected from prolonged light exposure, and is stable for 12 months from date of receipt. It is intended for research use only.
Solubility values reported on the safety data sheet include ~1 mg/mL in ethanol, ~0.1 mg/mL in a 1:9 solution of DMF:PBS (pH 7.2), and ~25 mg/mL in DMF and DMSO. The substance is not classified as hazardous according to the Globally Harmonized System (GHS).
