Description

From STEMCELL Technologies, this small molecule is classified as a MEK/ERK pathway inhibitor and acts as a reversible, selective, and ATP-competitive inhibitor of B-RAF, with higher potency towards the V600E mutant B-RAF compared to wild-type B-RAF or C-RAF (IC₅₀ values of 0.8, 3.2, and 5 nM, respectively). It is also reported as a tyrosine kinase inhibitor that inhibits ABL, SRC, LCK, and YES, and is used to inhibit melanoma growth.

The compound has a chemical formula of C₂₃H₂₀F₃N₅O₂S₂, a molecular weight of 519.6 g/mol, and a purity of ≥ 98%. It appears as a crystalline solid with low solubility in aqueous media, and is soluble in DMSO at ≤ 55 mM and in absolute ethanol at ≤ 2 mM. The product is stable at -20°C as supplied and should be protected from prolonged exposure to light. For cell culture use, stock solution should be diluted into culture medium immediately before use, avoiding final DMSO concentration above 0.1% due to potential cell toxicity.

Published applications include cancer research, where the compound decreases ERK phosphorylation and inhibits cell proliferation in cells expressing the B-RAF V600E mutation, and prevents tumor growth in mouse xenograft models of B-RAF(V600E) melanoma and colon cancer. In disease modeling, it has been shown to alleviate acetaminophen-induced liver injury through nanomolar inhibition of receptor-interacting protein (RIP3) in mice.
