Description

A selective, potent, and cell-permeant small-molecule inhibitor from STEMCELL Technologies, this compound targets casein kinase 1 (CK1) and the Activin/NODAL/TGF-β signaling pathway. It inhibits CK1 and CK1δ from S. pombe with IC₅₀ values of 200 nM and 300 nM, respectively, and the transforming growth factor β1 (TGF-β1) type I receptor (ALK5) with an IC₅₀ of 500 nM. The compound is also reported to inhibit p38 MAPK. Also known as Casein Kinase I Inhibitor, it has the chemical name 4-[4-(2,3-dihydro-1,4-benzodioxin-6-yl)-5-(2-pyridinyl)1H-imidazol-2-yl]-benzamide, molecular formula C₂₃H₁₈N₄O₃, molecular weight 398.4 g/mol, and CAS number 301836-43-1.

Supplied as a crystalline solid at ≥ 98% purity, the product is stable at -20°C as supplied for 12 months from the date of receipt when protected from prolonged light exposure; for long-term storage, a desiccant is recommended. Approximate solubilities are ~0.1 mg/mL in 1:10 DMSO:PBS (pH 7.2), ~10 mg/mL in ethanol, and ~30 mg/mL in DMSO and DMF. Stock solutions in DMSO are stable at concentrations up to 75 mM, and up to 25 mM in absolute ethanol. The compound is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS).

Published applications include reprogramming of mouse somatic cells into pluripotent cells, where the compound acts as a chemical substitute for OCT4 alongside Forskolin and 2-methyl-5-hydroxytryptamine (Hou et al., 2013), and cancer research, including induction of apoptosis in multiple myeloma cell lines (Hu et al., 2015) and selective anti-leukemic activity against leukemia stem cells versus normal hematopoietic stem and progenitor cells (Järås et al., 2014).
