Cl-Amidine hydrochloride is a cell-permeable, irreversible pan-peptidylarginine deiminase (PAD) inhibitor supplied as 1 mg of lyophilized powder for research use. This small-molecule probe targets PAD1, PAD2, PAD3, and PAD4 enzymes, which catalyze the post-translational conversion of protein arginine residues into citrulline. By suppressing PAD activity, Cl-Amidine enables researchers to investigate citrullination-dependent mechanisms in vitro and in vivo, including neutrophil extracellular trap (NET) formation, chromatin decondensation, and the autoimmune pathways implicated in rheumatoid arthritis, lupus, and multiple sclerosis.

Working concentrations typically fall in the 1 to 200 µM range, with reported IC50 values near 8 µM for PAD2 and sub-micromolar activity against PAD4 in biochemical assays. The compound dissolves readily in DMSO and can be diluted into aqueous buffers for cell culture treatment. Storage at -20°C in a desiccated vial preserves long-term stability, and stock solutions are commonly aliquoted to minimize repeated freeze-thaw cycles.

STEMCELL Technologies provides this inhibitor in single-use vials for academic and pharmaceutical laboratories studying citrullinome biology, cancer epigenetics, and inflammatory signaling pathways.
