Description

A reversible, cell-permeable WNT pathway activator from STEMCELL Technologies, this compound inhibits glycogen synthase kinase 3 (GSK3α and GSK3β). It demonstrates at least 500-fold selectivity for GSK-3 versus 20 other serine/threonine or tyrosine kinases (Ring et al.).

The product is supplied as a crystalline solid with a purity of ≥ 98% and a molecular weight of 486.3 g/mol (CAS 556813-39-9). Reported solubility is approximately 0.2 mg/mL in 1:4 DMSO:PBS (pH 7.2) and approximately 1 mg/mL in DMSO and DMF. According to manufacturer documentation, it affects the differentiation of induced pluripotent stem cells (iPSCs) and mesenchymal stem cells (MSCs), activates glycogen synthase in cells to lower blood glucose levels and improve glucose disposal in insulin-resistant rats and diabetic db/db mice (Ring et al.), and reduces tau phosphorylation in postnatal rat cortex and hippocampus in a model of Alzheimer's disease pathology (Selenica et al.).

The compound is stable at -20°C as supplied and should be protected from prolonged exposure to light. DMSO stock solutions should be prepared fresh, aliquoted to limit freeze-thaw cycles, and diluted into culture medium immediately before use, with final DMSO concentration kept at or below 0.1%. The substance is not classified as hazardous under GHS, but appropriate chemical-resistant gloves, lab coat, safety glasses, and respiratory protection are recommended during handling.
