Description

This small molecule from STEMCELL Technologies is a prototypical adamantyl arotinoid of the retinoid-related molecule family that acts as a selective agonist of the retinoic acid receptor (RAR). It is also known by the synonym AHPN. The chemical name is 6-(4-Hydroxy-3-tricyclo[3.3.1.13,7]dec-1-ylphenyl)-2-naphthalenecarboxylic acid, with CAS number 125316-60-1, molecular formula C27H26O3, and molecular weight 398.5 g/mol. The compound is supplied as a crystalline solid with a purity of ≥ 95%.

Reported binding affinities include a Kd of 6.5 µM for RAR α, 2.5 µM for RAR β, and 77 nM for RAR γ. Solubility values provided in the product documentation are ~0.05 mg/mL in a 1:1 solution of DMSO:PBS (pH 7.2), ~1 mg/mL in EtOH, ~16 mg/mL in DMSO, and ~10 mg/mL in DMF; alternative concentration-based solubility figures are ≤ 2.5 mM in absolute ethanol and ≤ 40 mM in DMSO. As an example, to prepare a 10 mM stock solution in DMSO, 1 mg is resuspended in 251 μL of DMSO. The product is stable at -20°C as supplied, should be protected from prolonged exposure to light, and remains stable as supplied for 12 months from the date of receipt. The compound has low solubility in aqueous media.

Published applications include increasing the speed and number of pre-induced pluripotent stem cell colonies generated from mouse embryonic fibroblasts transfected with Oct4, Sox2, c-Myc, and Klf4, and inducing cell cycle arrest and apoptosis in a variety of cancer cells, including melanoma, breast, lung, and prostate cancer cells. The compound is also reported to decrease mRNA expression of squamous differentiation markers cytokeratin 1, involucrin, and SPR1 in the human head and neck squamous cell carcinoma cell line UMSCC22B. It is classified as hazardous — specifically as Reproductive toxicity Category 2 under the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200), with the hazard statement "Suspected of damaging fertility or the unborn child." Products are for research use only and not intended for human or animal diagnostic or therapeutic uses unless otherwise stated.
